Levels of neurofilament subunits, potential biomarkers of motor axon breakdown, are increased in amyotrophic lateral sclerosis (ALS) patient's CSF but data on blood are not available. We measured blood levels of the phosphorylated axonal form of neurofilament H (pNF-H) by ELISA in transgenic rodent models of superoxide dismutase 1 (SOD1) ALS, and in 20 ALS patients and 20 similar aged controls monthly for 4 months. All symptomatic rodent ALS models showed robust levels of blood pNF-H, while control rodents or mice transgenic for unmutated SOD1 showed no detectable blood pNF-H. Average pNF-H levels in the G93A SOD1 mouse progressively increased from day 74 through death (day ∼130). Median blood pNF-H level in ALS patients was 2.8-fold higher than controls ( p  < 0.001). Median ALSFRS-R declined a median of 0.8 pt/month ( p  < 0.001); higher baseline pNF-H level appeared to be associated with faster ALSFRS-R decline over 4 months ( p  = 0.087). The median rate of decline in ALSFRS-R was 1.9 pt/month in patients with baseline pNF-H levels above the median pNF-H value of 0.53 ng/mL; ALSFRS-R declined at a median of 0.6 pt/month in patients below this level. The pNF-H levels were relatively stable month to month in individual patients, raising questions regarding the molecular pathogenesis of ALS. Baseline control human pNF-H levels were higher in men than women and increased minimally over time. These data suggest that blood pNF-H can be used to monitor axonal degeneration in ALS model rodents and support further study of this protein as a potential biomarker of disease prognosis in ALS patients.  相似文献

A mild degree of hemolytic anemia was induced in neonatal rats by a single subcutaneous injection with phenylhydrazine (PHZ). The ability to respond to this challenge was determined by monitoring serum erythropoietin (EP) levels at 6 and 12 hr. At 6 hr after PHZ, EP reached a level of 4.25 ± 1.03 U/ml, and at the 12-hr interval it had increased to 6.12 ± 1.49 U/ml. Based on these data, it is our suggestion that PHZ-induced hemolysis affords an effective stimulus for EP production in the neonatal rodent.  相似文献

A procedure has been developed for obtaining haemoglobin-free, erythrocyte ghosts under ionic conditions approximating that of the cell cytoplasm. Haemolysis was effected by incorporating glycol into cells suspended in the isoionic medium and then diluting with a large volume of glycol-free medium.The ghosts were of uniform spherical shape throughout the preparative procedure and were impermeable to macromolecules.Analysis of polypeptides by sodium dodecyl sulphate-gel elecrophoresis at each stage of preparation and comparison with ghosts prepared under hypo-ionic conditions served to distinguish membrane components from those of cytoplasm.  相似文献

The valve activity and water loss from aerially exposed Mytilus edulis L. and Modiolus modiolus (L.) have been investigated in a series of laboratory experiments. Mytilus is capable of maintaining long periods of complete, or almost complete, valve closure when exposed in air, and this allows the retention of water in the mantle cavity and protects the tissues from evaporative water loss. Over periods of three days or more the amount of water lost from emersed Mytilus was found to be less than that retained in the mantle cavity at the beginning of exposure, suggesting that during normal periods of exposure the tissues are never directly subject to water loss. In contrast, Modiolus shows periods of gaping during which water loss is rapid due to drainage from the mantle cavity and evaporation from the tissues. The exposure of the tissues to air that results from gaping, makes the water loss susceptible to environmental influences of which wind was found to be the factor which caused the greatest increase in water loss and, as a consequence, an important factor in causing death through dehydration. The different abilities of Mytilus edulis and Modiolus modiolus to control water loss may be related to their intertidal distributions.  相似文献
